Raw garlic's component, diallyl disulphide (DADS), may prevent hypertrophy by preventing the production of H2S and/or mitochondria.

Isoproterenol was administered subcutaneously for 14 days at a dose of 5 mg/kg/day to rats in order to produce cardiac hypertrophy. The third, fourth, and fifth groups of rats were fed aqueous garlic homogenate, DADS, and NaHS (liberate H2S) for 14 days at doses of 250 mg/kg/day, 50 mg/kg/day, and 14 M/kg/day, respectively. Garlic and DADS normalised mitochondrial ETC-complex activities, mitochondrial enzyme activites, and mitochondrial biogenetic and apoptotic gene expression while decreasing heart hypertrophy indicators. In addition to raising NRF2 protein levels and Tfam gene expression, garlic and DADS raised rat cardiac eNOS and p-AKT levels. NaHS, which produces H2S in vivo, was administered, however normalisation was not noticed. In conclusion, the combination of garlic and DADS stimulates mitochondrial biogenesis and reduces heart hypertrophy by rats' eNOS-Nrf2-Tfam pathway.

Thus, Diallyl disulfide reduces the isoproterenol-induced cardiac hypertrophy in rats through stimulating the eNOS-Nrf2-Tfam pathway.
