What type of plasmids are present?
Plasmids generally exist in 3 conformations: super coiled, open circular and linear. In-vivo, plasmid DNA is a tightly super coiled circle to enable it to fit inside the cell. In the laboratory, when we extract plasmid , most of the DNA will remain super coiled, but a certain amount will be in open circular and linear conformations. The open circular form of plasmid exist due to the break in only one of the strands, the DNA will exist as circular. The break will allow rotation around the phosphodiester back bone of DNA due to which super coils will be released, thus creating open circular DNA. The plasmid exists in the linear form when there is a cut in the circular DNA.
